Reflectance confocal microscopy (RCM) images tissue in its native state, i.e. without the use of exogenous contrast agent/dye. Laser scanning confocal microscopy (b) focuses a single point of laser light through a small aperture (pinhole) and scans sequentially across the sample point by point. Spinning disk confocal microscopy (c) illuminates the sample with a rotating pattern of 1,000's of pinholes for complete simultaneous confocal illumination.
So images have better contrast and are less hazy.

Its fundamental purpose is to provide so-called optical sectioning and thus to enable the recording of three-dimensional images, which is impossible to achieve with conventional wide-field microscopy. Confocal microscopy offers several advantages over conventional widefield optical microscopy, including the ability to control depth of field, elimination or reduction of background information away from the focal plane (that leads to image degradation), and the capability to collect serial optical sections from thick specimens.
In the most widely used and simple form of the instrument, laser light is focused by an objective lens (Fig. 21.1) on to the object, and the reflected, or fluorescent, light focused on to a photodetector via a beamsplitter. An image is built up by scanning the focused spot relative to the object, and is usually stored in a computer imaging system for subsequent display. Laser scanning confocal microscopy, more generally referred as confocal microscopy, is an established microscopy technique that allows obtaining 2D or 3D high-resolution images of relatively thick samples. Confocal microscopy is a powerful technique to image fluorescent samples given its high efficiency, low background noise and the capability to optically section samples into different focal planes.
