Confocal microscopy offers several advantages over conventional widefield optical microscopy, including the ability to control depth of field, elimination or reduction of background information away from the focal plane (that leads to image degradation), and the capability to collect serial optical sections from thick specimens.

In the most widely used and simple form of the instrument, laser light is focused by an objective lens (Fig. 21.1) on to the object, and the reflected, or fluorescent, light focused on to a photodetector via a beamsplitter. An image is built up by scanning the focused spot relative to the object, and is usually stored in a computer imaging system for subsequent display.
